In our Protocol, we described how to perform the fluorescence imaging of plant salt stress. We did not, however, mention some logistical considerations related to imaging multiple plants. To improve efficiency and reproducibility, the three plants for each experiment were imaged simultaneously. To prepare the images for publication, the photographs were cropped so that each image contained a single complete plant. Inevitably, there was some overlap, with some “crops” including parts of other plants. If bits of other plants were present in the cropped image, we covered these using a shape with a black background. All these image processing steps were carried out in PowerPoint V2019.

This process was applied to Figure 8 and Supplementary Fig. 16 of the published article. Figure 1 below shows the unedited version of Fig. 8. The differences can be seen in the top right corner of all images in the first column (0 mM) and right center of all images in the second column (50 mM).

The unedited version of Supplementary Fig. 16 can be found in the Supplementary Information to this addendum. The differences can be seen in the top right corner of all images in column 7 (50 μM) and left center of all images in column 8 (100 μM).

The Supplementary Information also contains the original versions of two other figures in which the laser confocal’s own scale bar was covered if it was present in a distracting position:

  • Supplementary Figure 5: bottom right/left corners of all images in the first and third rows.

  • Supplementary Figure 9: top right/top left/bottom right corners of all images in the first and third rows.
