Rosiglitazone and trametinib exhibit potent anti-tumor activity in a mouse model of muscle invasive bladder cancer

New treatments are needed for muscle invasive bladder cancers. Here, the authors show that combined Pparg activation and MEK inhibition using FDA approved drugs shrinks tumor volume and induces a Basal/Squamous-to-Luminal shift in the urothelium as well as in invading tumors.

Modulating the PPARγ pathway upregulates NECTIN4 and enhances chimeric antigen receptor (CAR) T cell therapy in bladder cancer

Enfortumab vedotin (EV) is the current standard treatment for advanced bladder cancer, but resistance typically develops within a year, highlighting the need for new therapies. This study demonstrates that NECTIN4-targeting CAR T cells are effective against bladder cancer, including EV-resistant cells, and their potency can be further enhanced by using rosiglitazone to boost NECTIN4 expression.

Asc-1 regulates white versus beige adipocyte fate in a subcutaneous stromal cell population

Adipose tissue is composed of a variety of cell types, including adipocyte precursor populations, that contribute to adipose tissue function upon differentiation. Here, using scRNA-sequencing of adolescent and adult mouse subcutaneous adipose tissue, the authors identify an Asc-1 positive preadipocyte population that is enriched in adolescent subcutaneous fat and demonstrate that loss of Asc-1 triggers spontaneous beige adipocyte differentiation.

Brain PPAR-γ promotes obesity and is required for the insulin–sensitizing effect of thiazolidinediones

Although the use of thiazolidinediones in the treatment of type 2 diabetes is controversial, they still comprise an important class of insulin-sensitizing drugs. In two reports, new evidence emerges that these drugs rely on the activation of their target, PPAR-γ, in the brain to improve hepatic insulin sensitivity and increase food intake and reduce energy expenditure.

Gerhard Krönke and his colleagues show that PPARβ/δ regulates Wnt signaling in osteoblasts, which in turn determines the proper ratio of OPG to RANKL and thus bone homeostasis. They go on to show that pharmacological activation of this receptor normalizes bone density in a mouse model of osteoporosis.
