Harnessing an integrated glyco-nanovaccine technology for enhanced cancer immunotherapy

Niimura et al. investigates the potential of an integrated glyco-nanovaccine (iGN) comprising gold nanoparticles conjugated with a synthetic Toll-like receptor 7 ligand, sugar chains, and peptide antigens for cancer immunotherapy. Treatment with iGN increases tumor-infiltrating CD8+ T cells and inhibits tumor growth.

Assembly and activation of the Hippo signalome by FAT1 tumor suppressor

Dysregulation of the Hippo signaling is a frequent event in human malignancies, but the molecular mechanisms remain unclear. MAP4K family kinases act in parallel to MST1/2 to activate LATS1/2 in the Hippo pathway

A variety of signals have been reported to either activate or inhibit the Hippo kinase cascade. Here, Meng et al. show that mitogen activated protein kinase kinase kinase kinase (MAP4K) family members function in parallel to and are partially redundant with MST1/2 in regulating LATS in response to upstream signals.

Pharmacological inhibition of Mint3 attenuates tumour growth, metastasis, and endotoxic shock

Sakomoto et al. identify naphthofluorescein as a mint3 inhibitor that disrupts the Mint3–FIH-1 interaction and attenuates HIF-1 activity. In vivo experiments in mice reveal a reduction in tumor growth with attenuated inflammatory cytokine production and endotoxic shock, presenting an option for targeted therapies for cancer and inflammatory diseases that avoid severe adverse effects.
