From: Structural and functional characterization of a DARPin which inhibits Ras nucleotide exchange
Parental HCT116 cells and lentivirally transduced inducible K27 and K55 HCT116 cells were grown in a soft agar colony formation assay in the presence and absence of 1 μg ml−1 doxycycline for 14 days. Anchorage-independent growth was measured by alamar blue assay fluorescence and for each cell line was normalized to its respective untreated control (n=4). The statistical significance of the difference in growth signal observed in response to the addition of doxycycline was calculated by ANOVA followed by t-test adjusted for multiple comparisons using Bonferroni correction where **P<0.01 and ***P<0.001.
