A Schema of an in vivo screening strategy for identifying SENPs that modulate PDAC metastasis. Lentivirus particles targeting each SENP family member were individually produced in HEK293T cells and subsequently infected into GFP and luciferase-producing PDAC cell lines (PANC-1 and Patu-8988t). Infected cells were selected with puromycin and then mixed (for each SENP-targeted cell line 3 × 104 cells were employed) for orthotopic injection into pancreas of ten nude mice for each cell line. Eight weeks later, vigorous metastatic liver nodules detected with the IVIS imaging system were isolated from tumor-bearing mice. GFP-positive metastatic cells were isolated with FACS sorting following single-cell suspension collection by liver digestion. Genomic DNA extracted from sorted cells was amplified and verified by DNA sequencing.
