Single-molecule FRET intensity distributions. The distribution of single particle spot intensities is described well by a single log-normal distribution for a the predominantly monomeric donor spots (n = 209,712), and c for acceptor spots from a monomeric doubly labelled (TM1–TM4, Cy3–Cy5 distance ~1.5 nm) NTS1 control sample (n = 2816), while for b the FRET acceptor dimer spots it was described by the sum of two log-normal distributions representing high (dotted line) and low (dashed line) FRET dimeric conformations (n = 8561). Spot lifetime (d–f) was similar for all populations indicating dimer lifetime on the order of or greater than the photobleaching timescales.
