Kinetic model of the NCBD-ACTR interaction. The two subpopulations of NCBD differ by the cis/trans isomers of the peptide bond involving Pro20, which are interconverting slowly; both bind ACTR, but with different association rate coefficients. The two complexes can be distinguished experimentally by their dissociation rate coefficient, which is greater for the NCBDP20,cis-ACTR complex. The difference in association and dissociation rates translates into an eightfold higher affinity of the NCBDP20,trans-ACTR complex
