SHQ1 depletion impedes leukemogenesis in NOTCH1-induced T-ALL model. a Graphical illustration of NOTCH1-induced T-ALL mouse model. Fetal liver cells were infected with MSCV retrovirus, enabling simultaneously expression of ICN1 and shRNA targeting non-specific control (shCtrl) or murine SHQ1 (shmSHQ1). Two million GFP+ transduced cells were intravenously injected into each irradiated recipient, followed by assessment of leukemia cell dissemination. b Kaplan–Meier survival curves of fetal liver cell transplants expressing vector (black line, n = 5), ICN1/shCtrl (blue line, n = 5), or ICN1/shmSHQ1 (red line, n = 5). The p value was determined by log-rank test. Around day 55 post transplantation, ICN1/shCtrl mice became moribund, then three mice in each group were killed for assessment of T-ALL in vivo. c Representative flow cytometry analysis of GFP+ and CD4+CD8+ leukemia cell distribution in each group. d Representative spleen and bone images in each transplant. e Representative histological images of murine SHQ1 and PCNA in spleen sections from each group. Scale bar, 50 μm
