  1. Respective ligands were titrated into the GluN1a–GluN2B ATD heterodimer. Superscript indicates the pH at which the experiments were performed to generate the given value. pH-boost is the ratio of KDpH 7.6 to KDpH 6.5. The isolated GluN1a–GluN2B ATD expressed in HEK cells was used for all ITC experiments, except for one set of experiments in which we titrated ifenprodil into the isolated GluN1b–GluN2B ATD used for crystallization. Values are presented as mean ± S.E.M. All experiments were performed in triplicate as described in Methods. Side chain volumes were calculated using MolInspiration (molinspiration.com). n/a not applicable
