From: Adipose group 1 innate lymphoid cells promote adipose tissue fibrosis and diabetes in obesity

Fig. 3

Adipose ILC1s contribute to obesity-associated adipose fibrosis. Briefly, C57BL/6 mice were fed a HFD for 3 weeks, and 5 × 104 adipose ILC1s were sorted (>95% purity) from adipose tissue and adoptively transferred intravenously (i.v.) into Prkdc−/−IL2rg−/− mice. The recipient mice were fed a HFD for 4 weeks. a Schematic of the experiment. b Representative plots indicate adoptively transferred adipose ILC1s in the VAT of recipient mice 4 weeks after transfer. c Body weight. d Glucose tolerance test. e Expression levels of fibrosis-related genes, assessed by q-PCR. f Representative Masson’s trichrome C and Sirius red staining in VAT of mice with different treatment (scale bar: 100 μm). g, h Representative αSMA staining (scale bar: 100 μm) and quantification of αSMA-positive area in VAT of mice with different treatment. Arrows indicate positive staining areas. *P < 0.05 vs. CHOW group; **P < 0.01 vs. CHOW group; ##P < 0.01 vs. HFD+PBS group (two-way ANOVA tests followed by Bonferroni post hoc test). The data are representative of three independent experiments, with n = 4–6 mice per group. Error bars indicate s.d.
