Threonine but not methionine deficiency leads to lysine overload. To confirm that threonine deficiency alone is responsible for accumulation of lysine and intermediates, the endo-metabolome of the threonine auxotroph Mtb ∆thrB was determined and compared to Mtb ∆thrA. Accumulation of DAP, lysine and n-acetyl-lysine was detected in both strains. Only threonine but not methionine reversed lysine accumulation in Mtb ∆thrA. All values are the average of three biological replicates (n = 3) ± s.d. (error bars depict standard deviation) and representative of two independent experiments. **p-value < 0.01 in Student's t-test
