Tractions forces applied via lectin-coated beads activate the β2AR. a Neutravidin-agarose beads (1 µm) were coated with biotinylated WGA or control dolichos biflorus agglutinin (DBA) as described in Methods. The latter lectin has specificity for α-linked N-acetylgalactosamine. They were then let to sediment for 1 h on HEK-293 cells expressing the indicated YFP-tagged wt of mutant receptors, with or without exogenous β-arrestins (β-arrs), in 2 cm2 well plates before 15 min of orbital rotation at 100 r.p.m. Cells were fixed and labeled with anti-ezrin antibodies.
