Following vesicle docking (I; for an enlarged image please see Fig. S12) local point contact is established between the vesicles (II), leading towards membrane mixing and the emergence of hemifusion diaphragm (III). While the latter is expanding (IV), membrane thinning (shown with white arrows, enlarged in Fig. S13) can be observed at the juncture, indicating the location of the eventual pore opening (V). Such position of fusion pore was previously observed in simulations49. Slight lateral deformations in the newly fused vesicles along with less frequent inclusions in the lumen are likely remnants of the hemifusion diaphragm (VI). Scale bars represent 30 nm.
