Summary of the DSB-Spectrum variants described in this manuscript. The frequently used DR-GFP reporter is shown for comparison purposes. Each panel shows, from top to bottom (1) the fluorescence changes caused by repair of the site-specific DSB in the depicted reporter construct, (2) a cartoon example of a population after reporter activation, in presence or absence of DNA-PKcs inhibition as an example of a DSB-repair manipulation strategy, (3) quantification of the results in panel 2, and (4) the model resulting from the data obtained with the reporter construct.
