AC BEAS-2B cells were infected with Spn D39, WT or Δply for 16 h. Gene expression was determined by qPCR. DF BEAS-2B cells were treated with pneumolysin in indicated concentrations for 16 h. Expression of IL-8, NAMPT and NMNAT1 was determined as indicated. GI BEAS-2B cells were treated with 120 nM inactive pneumolysin for 16 h. Expression of IL-8, NAMPT and NMNAT1 was determined as indicated. Statistics: two-tailed paired t-test (AC, GI); sigmoidal dose-response fit with variable slope (DF); Significances were determined against wild-type infected (AC) or unstimulated controls; N = 4 biologically distinct samples (AC) 3 biologically distinct samples (DI); box plots: line at mean, box ranges from min to max; results are normalized against untreated controls. Ply pneumolysin.
