  1. Fluorescence observed by TMR labeling of one or two cysteine residues inserted into the three-helix bundle of monomers (MDIns2/IQ/3HB) and zippered dimers. Fluorescence was analyzed by a ratio of the emission values to that of the absorption values for each construct from four independent measurements (n = 4). Mean values (±SD) are reported. The molar ratio was calculated by comparing the myosin concentration to the concentration of the incorporated TMR.
