(1): The MNAzyme will self-assemble on-site by consuming miRNA-21 and Ca2+ ions and silencing HSP70 mRNA before photothermal therapy (PTT) treatment. (2) Silencing miR-21 causes Phosphatase and TENsin homolog deleted on chromosome 10 (PTEN) upregulation for PTT sensitization. (3) Ca2+ will be absorbed by mitochondria, and curcumin will promote the further release of Ca2+ from the ER and inhibit the Ca2+ efflux pump. Mitochondrial calcification can reduce HSP-chaperones adenosine triphosphate (ATP) production. (4): IR780 will target mitochondria to achieve precise mild PTT treatment and produce ROS-mediated photodynamic therapy.
