Aberrant TWEAK/Fn14 signalling in TNBCs trigger NAMPT SE activation to augment NAMPT expression via chromatin looping. MAFG binds to the TWEAK-driven SE to regulate its activity which in turn leads to NAD+ and ATP metabolic rewiring, resulting in enhanced TNBC proliferation, filopodia formation and metastasis.
