Ratio of the plasmid-containing cells to the total number of cells in CFU/mL of E. coli MG1655 with the non-replicative plasmid pAM34 which bears an ampicillin resistance cassette. The total cell density and density of cells bearing the plasmid were measured every 10 days for 30 days of four independent biological replicas for four conditions: i) minimal media with CHL (chloramphenicol), ii) minimal media with AZM (azithromycin), iii) minimal media with ampicillin– maximal plasmid retention, iv) minimal media with no antibiotic– maximal plasmid loss. In the absence of CHL or AZM the initial cultures can only grow two logs until arriving to a maximum stationary phase, explaining the maximum loss of two log in the absence of ampicillin.
