From: A synthetic chronogenetic gene circuit for programmed circadian drug delivery

A therapeutic chronogenetic circuit was created and transduced into murine induced pluripotent stem cells (miPSCs) using lentivirus. These miPSCs were then differentiated into tissue engineered cartilage. The circuit was driven by the Per2 promoter, activated by core clock component Per2 and produced both a luciferase reporter and the therapeutic drug, IL-1Ra. The Per2-IL1Ra:Luc circuit is capable of delivering an anti-inflammatory drug to target inflammatory flares.
