Derangement of the Hippo signaling pathway components lead to the activation of YAP-TEAD activity in the nucleus leading to increased gene expression of matrix molecules and modifying enzymes (such as LOX) leading to increased secretion of LOX from the fibrotic alveolar epithelial cells, which contributes to increased crosslinking of secreted matrix in the ECM. This increase in matrix crosslinking causes further changes in the mechanical stiffness that leads to further increase in YAP activity. Created in BioRender. Wagner, D.
