A549 cell cultures were treated individually with A the alkyne reagent 1 and then clicked to Azide-Cy5 (B) the azide reagent 2 and then clicked to DBCO-Cy3. For quantitating crosslinking, A549 cell cultures were treated with both precursors, then coupled with DBCO-Cy3 either C before or D after precursors were clicked together through click-linking. Fluorescence signal is reduced after clicking and is correlated to lower azide availability due to crosslink formation (see methods). Source data are provided as a Source Data file.
