For a and b cells were co-transfected with the indicated combinations of the IL-17RA, IL-17RB, and ACT1 constructs. For c and d cells were first transfected with siRNA-1 (Supplementary Fig. 6c) to knock down endogenous IL-17RA expression and then transfected with indicated plasmids. Cells were stimulated with IL-25 at 25 nM. IL-6 levels in the medium were measured with ELISA. Each circle represents the result from one of three independent experiments. The bars represent means and standard deviations. P values were calculated with two-tailed Student’s T-test.
