The peak at EFRET ~0 corresponds to AF488-conjugated bound/unbound DNA. (Right panels) Corresponding single-molecule photon bursts using PIE excitation (donor channel, green; acceptor channel, red). Highlighted gray areas indicate coincidence between donor and acceptor signals but not necessarily FRET between the bursts. From time-gating FRET analysis, intermolecular FRET efficiencies were calculated. Similar results were obtained from 2 independent experiments.
