YAP/TAZ get a STING in the tail

Ageing involves numerous tissue alterations, such as cell senescence, inflammation and changes in tissue mechanics. How these different processes are linked is poorly understood. Sladitschek-Martens now show that reduced activity of key cellular mechanosensors, transcription co-activators YAP/TAZ, is an important driver of ageing of stromal and contractile cells, leading to cell senescence.

Investigating the status of YAP/TAZ in mouse tissues, the authors noted that their transcriptional activity declines with ageing in skin fibroblasts (a primary stromal cell type in the skin) and in contractile cells, including in vascular smooth muscle cells and cardiomyocytes. Deletion of YAP/TAZ or perturbed extracellular matrix mechanosignalling in young mice induced premature ageing of the skin and the aorta, whereas ectopic expression of YAP in fibroblasts was sufficient to prevent several features of skin ageing.
