Targeting IL-1Ra to attenuate fungal infection

IL-1 receptor antagonist (IL-1Ra) secreted by macrophages might be a therapeutic target in invasive fungal sepsis, according to a study by Stefan Freigang and colleagues.

Candida-infected mice with a conditional deletion of Il1rn in macrophages had lower kidney fungal titres and inflammation than controls and were able to control the infection by day 7. This protective effect of IL-1Ra deletion seemed to be driven by enhanced recruitment of neutrophils with high antifungal capacity to the kidneys and was associated with an attenuated hyperinflammatory response. Administration of recombinant IL-1Ra to these transgenic animals, which restored circulating but not tissue levels of IL-1Ra, abrogated the renoprotective effects of IL-1Ra deletion.
