Fitting parameters of FIDA binding assays of (NIAU)2 titrated into FXNALC or FDX2ALC, and competition assays of unlabelled FDX2 or FXN titrated into a preformed complex of FXNALC or FDX2ALC with the (NIAU)2 complex. FXNALC Rh and FDX2ALC Rh corresponds to the hydrodynamic radius of the labelled proteins. Complex Rh corresponds to the hydrodynamic radius of the complex formed between (NIAU)2 and FXNALC or FDX2ALC, as indicated. For binding assays, 20 nM FXNALC or FDX2ALC was titrated with 0–8 µM (NIAU)2 or 0–64 µM (NIAU)2, respectively. For competition assays, 20 nM FXNALC was incubated with 1 µM (NIAU)2 and titrated with 0–64 µM unlabelled FDX2, and 20 nM FDX2ALC was incubated with 8 µM (NIAU)2 and titrated with 0–256 µM unlabelled FXN. The mathematical model used to fit competitive binding assays is derived in Supplementary Equation 1. Fixing the Kd of FXN in the competition assay of FXN into FDX2ALC-(NIAU)2 did not refine the fit of the data and was thus left as a floating parameter. (Parameters are calculated on the basis of data in Fig. 2a–d).
