a, Experimental approach: mice were transcardially perfused following the pan-ExM-t protocol (Methods), brains were extracted and sliced. Brain slices were divided in half; respective halves were prepared for electron microscopy (EM) and pan-ExM-t, respectively. b,c Comparison of EM (b) and pan-ExM-t (c) images for equivalent brain tissue regions taken from the same brain slice. Orange arrows indicate synaptic clefts. d,e, EM images of brain tissue samples from mouse neocortex. f, g, h, EM images of brain tissue samples from cerebellum. Please note the preservation of subcellular features such as cellular membranes, synaptic vesicles, mitochondrial cristae, and Golgi apparatus. These results suggest that pan-ExM-t based perfusion protocols are sufficient to preserve these features.
