a Acetyl-CoA accumulation when using module 4b with different amounts of Mdh. More Mdh leads to accumulation of acetyl-CoA and a decrease in methylmalonyl-CoA (see Fig. 3). b Methylmalonyl-CoA and c acetyl-CoA accumulation when using module 4d with different amounts of CoA (1, 0.5 and 0.25 mM). 1 mM CoA produced the highest concentration of methylmalonyl-CoA and was thus chosen for subsequent experiments. All experiments were performed in technical triplicates and are displayed as mean ± SD.
