a, Cytometric bead arrays was performed to quantify CCL2 concentrations in sera isolated from uninfected (0) and 36 h infected (1.5) CTR and pDC-less mice. Each dot represents an individual. The data shown (mean +/− s.e.m.) are pooled from two independent experiments (n = 3/strain at 0, 7/strain at 1.5 p.i.).). An unpaired and nonparametric two-sided multiple t-test (Mann-Whitney) with Holm-Sidak method for correction was used for the statistical analysis. b, Weight curves of mice represented in Fig. 2a (n = 10 for each strain). c, Relative expression of Mx2 (data represented as mean +/− s.e.m.) in the spleens of C57BL/6 infected with MCMV at indicated time points (spleen, n = 3, except 2 at 2 days p.i.; lungs= 2, except 3 at day 0; liver=3, except 2 at 0 and 2 days p.i.). d, Expression levels of indicated genes was analyzed by RT-qPCR in indicated organs isolated from uninfected or MCMV-infected CTR, pDC-less and Ifnar1KO mice at indicated days after infection. Data were normalized to expression level of Actin gene. The data (mean + s.e.m.) shown are from two independent experiments (n = 4 for each condition and time point, except 3 pDC-less at 1.5 and 2 days p.i. and 3 Ifnar1KO at 2 days p.i.). Statistical analysis was performed as described in Fig. 2d. e, CD86 MFI values on indicated cDC subsets (mean +/− s.e.m.) were determined in CTR and pDC-less mice at indicated days after MCMV infection (n = 3 at 0, 8 for 1.5, 7 at 2 and 3 days after infection for both strains). Statistical analysis was performed as described in Fig. 2e.
