Example of ADCD gating strategy: Conjugated beads are gated based on Forward Scatter and Violet Side Scatter then presented on an APC-histogram. The APC peak is gated and displayed on a FITC-histogram which represents deposition of C3b/iC3b. The median fluorescence intensity (MFI) of the complement only, no antibody control (complement only) is subtracted from the test sample (sample) to give the antibody-dependent MFI.
