Plasma and brain concentrations after a single per os administration of AEF0117 in male and female mice (n = 3 per sex and per time point, data for males and females were averaged) after different doses of AEF0117 (a) 0.3 mg/kg; (b) 4 mg/kg and (c) 10 mg/kg and (d) in male rats after 1.6 mg/kg of AEF0117 (n = 3 per time point). Plasma concentration of AEF0117 from the multiple ascending dose (MAD) study in healthy volunteers after: (e) the first dose (due to severe weather conditions, there was no sample available in the cohort of AEF0117 2 mg at 24 h); and (f) the seventh and last dose (n = 5 for 0.6 mg; n = 6 for the other dose cohorts).
