a. Representative FACS plots of cell suspensions for 10x single-cell analysis (top figure panel). Cells were gated i. on singlets followed by ii. collection of DAPI- and CD45+ cells. The middle figure panel shows the gating strategy for single-nucleus RNA sequencing. The bottom figure panel shows the gating strategy for single-nucleus fixed RNA profiling. Cells were sorted into 1.5 ml tubes. b. For the enrichment of CD206+ cells, i. singlets were selected followed by ii. exclusion of DAPI + CD3 + CD19 + CD20+ cells. iii-iv-.
