STXBP5 regulated progerin expression. (A) Overexpression of lamin A and progerin via the pCAG-3HA plasmid. HA-tagged lamin A and HA-tagged progerin were detected with an anti-HA antibody. HEK293 wild-type (WT) cells was transfected with the empty vector pCAG-3HA were used as the control group (Ctrl OE). The data were normalized to \(\beta\)-Actin. (B) Overexpression of HA-Lamin A in the HEK293 cell line increased the lamin A transcript levels. Expression of HA-Progerin in the HEK293 cell line increased the lamin A and progerin transcript levels. Lamin A and progerin transcript levels were measured via qRT-PCR. (C) HA-progerin-expressing HEK293 cells were treated with STXBP5 sh1 or STXBP5 sh2 for the knockdown of STXBP5 or with shLuci as the control shRNA. Lamin A, progerin and STXBP5 transcript levels were measured via qRT-PCR. (D) Western blots revealed that the knockdown of STXBP5 led to reductions in the HA-tagged progerin protein level, and the bar graph shows the changes in HA-tagged progerin expression in three independent experiments. The orange arrowheads indicate progerin and Lamin A bands.
