(a–c) HUVECs transfected with control or Smad6 siRNA were subjected to FSS at indicated magnitudes for 12 h. Smad6 knockdown efficiency was confirmed by Q-PCR (a), n = 3 experiments, data are presented as means ± s.e.m. Cells were fixed and stained for Smad1. Nucleus/Cytoplasm intensity ratio of Smad1 was quantified, n = 60 cells for each group from 3 experiments. (d, e). Immunostaining of phospo-Smad1/5. Nucleus intensity of Smad1/5 was quantified (normalized to siCtrl 3 dyn/cm2 group), n = 60 cells for each group from 3 experiments, data showing all points from min to max. Scale bar: 25 μm.
