TGF-β builds a dual immune barrier in colorectal cancer by impairing T cell recruitment and instructing immunosuppressive SPP1⁺ macrophages

Targeting TGFBR1 in transplantable mouse colorectal tumor organoids improves response to anti-PD-L1 therapy. Mechanistically, TGF-β abrogates clonal expansion of T effector and memory phenotypes and indirectly inhibits T cell activity by regulating immunosuppressive tumor-associated macrophage activity.

Inherited defects of piRNA biogenesis cause transposon de-repression, impaired spermatogenesis, and human male infertility

piRNAs are small RNA molecules found primarily in the testes of mice and men. Stallmeyer et al. demonstrate that variants in specific genes can disrupt the formation of piRNAs, impairing spermatogenesis and causing human male infertility.

Stromal gene expression defines poor-prognosis subtypes in colorectal cancer

Eduard Batlle and colleagues find that poor-outcome subgroups of colorectal cancer are driven by genes expressed in tumor-associated stromal cells. They show that TGF-β expression is a common feature of poor-prognosis subtypes and demonstrate the use of anti–TGF-β molecules in patient-derived tumoroids and xenografts.
