Genetic variations interact with polybrominated diphenyl ether exposure to alter lipid homeostasis

Polybrominated diphenyl ethers (PBDEs) are linked to dyslipidemia, but why individuals respond differently is unclear. Here, the authors use a national biomonitoring cohort to reveal how specific genetic variants interact with PBDE exposure to increase risk, identifying a key mechanism that disrupts cholesterol metabolism.

Functional role of Tet-mediated RNA hydroxymethylcytosine in mouse ES cells and during differentiation

TET mediated RNA-hydroxymethylation (5hmC) has been detected in mammals, but its physiological role remains unclear. Here the authors map 5hmC during embryonic stem cell (ESC) differentiation and find that Tet-mediated RNA hydroxymethylation reduces the stability of crucial pluripotency related transcripts.

A selective USP1–UAF1 inhibitor links deubiquitination to DNA damage responses

Deubiquitinases (DUBs) are peptidases that remove ubiquitin from post-translationally modified proteins. The identification of a selective small-molecule inhibitor of the USP1–UAF1 deubiquitination complex reveals a role for deubiquitination in regulating the DNA damage response.

A quantitative assay for assessing the effects of DNA lesions on transcription

DNA damage products influence DNA replication but also may induce stalling or mutagenesis during transcription. A competitive transcription and adduct bypass assay provides a new approach for assessing the transcriptional effects of DNA lesions and links transcriptional arrest of several lesions to nucleotide excision repair pathways.
