Lactate released by tumor cells has been shown to inhibit antitumor NK cell responses. Here the authors show a mechanism for this effect in which lactate drives lysine lactylation of NK cells. A combination treatment also limits this lactylation to prevent NK cell dysfunction, thereby restoring antitumor immunity.
