The landscape and regulatory potential of eccDNAs in mammalian preimplantation embryos

Extrachromosomal circular DNA (eccDNA) is a regulatory element involved in genomic stability and gene regulation. This study identifies widespread eccDNA in early mammalian embryos. These eccDNAs arise from active genomic regions, increase during a major wave of gene activation, and can regulate key developmental genes.

Mechanism for DPY30 and ASH2L intrinsically disordered regions to modulate the MLL/SET1 activity on chromatin

Regulation of the MLL family of histone H3K4 methyltransferases on the nucleosome core particle (NCP) remains largely unknown. Here the authors show that intrinsically disordered regions of ASH2L and DPY30 restrict the rotational dynamics of MLL1 on the NCP, allowing more efficient enzyme-substrate engagement and higher H3K4 trimethylation activity.

Quiescence enables unrestricted cell fate in naive embryonic stem cells

Stem cell quiescence is generally considered as an inactive state with poised potential. Here, Khoa et al. find that quiescent embryonic stem cells actively maintain a dynamic reservoir of cells with unrestricted cell fate that converges on S-adenosylmethionine and H3K27me3 status.

Discovery of first-in-class inhibitors of ASH1L histone methyltransferase with anti-leukemic activity

The histone methyltransferase ASH1L plays a role in various diseases, including cancer, and has been validated as a therapeutic target; however, no inhibitors of ASH1L have been reported. Here the authors present small molecule inhibitors of ASH1L and demonstrate their on-target activity in leukemia cells and a mouse model of leukemia.
