Rab41-mediated ESCRT machinery repairs membrane rupture by a bacterial toxin in xenophagy

Xenophagy targets cytosolic bacteria, but their membrane homeostatic system is not known. Here the authors show that the ESCRT machinery repairs xenophagic membrane damaged by a bacterial toxin, which is regulated by the TOM1L2–Rab41 that recruits VPS4.

TBC1D9 regulates TBK1 activation through Ca²⁺ signaling in selective autophagy

Xenophage, or autophagy directed against pathogens, might be involved in the clearance of bacterial infection, but the mechanism is unclear. Here, the authors show that ubiquitinated bacteria are bound by TBC1D9 via a Ca²⁺-dependent interaction to induce downstream activation of TBK1 signalling and xenophagy, as well as mitophagy.

Cap analogs with a hydrophobic photocleavable tag enable facile purification of fully capped mRNA with various cap structures

Removing immunogenic uncapped mRNA from transcribed mRNA can be challenging, but is critical in mRNA research and clinical applications such as vaccines. Here, authors develop hydrophobic photocaged tag-modified cap analogs, which can be used to separate capped mRNA from uncapped mRNA, with subsequent tag removal using photo-irradiation.

ARID2 is a pomalidomide-dependent CRL4^CRBN substrate in multiple myeloma cells

Immunomodulatory drugs are used for the treatment of multiple myeloma. ARID2, a component of the PBAF chromatin-remodeling complex, is a new pomalidomide-induced neosubstrate of CRL4^CRBN, which accounts for its superior efficacy over lenalidomide.

Large-scale analysis of small molecule-RNA interactions using multiplexed RNA structure libraries

Targeting RNA with small molecules represents an attractive medicinal approach, and profiling the small molecule-binding landscapes of various types of RNA structures is critical. Here, the authors develop a large-scale approach for investigating small molecule-RNA structure interaction profiles using a multiplexed pull-down assay with various RNA structure libraries, and validate the assay using G-clamp and thiazole orange derivatives.
