Rosiglitazone, identified through integrative network-based drug repurposing and Mendelian randomization, alleviates age-related sarcopenia in mice. It enhances muscle strength and mass, possibly through gut microbiota remodeling, activation of Igf1 signaling, and inhibition of muscle atrophy–related ubiquitin ligases.
