Histone H3K9 methylation is dispensable for Caenorhabditis elegans development but suppresses RNA:DNA hybrid-associated repeat instability

Susan Gasser and colleagues find that methylation at histone H3 lysine 9 (H3K9me) is required for repression of simple repeats and transposons in Caenorhabditis elegans. Loss of H3K9me in worms leads to extensive accumulation of insertions and deletions at repeat elements, which correlate with R-loop formation and increased sensitivity to replication stress.

Genetic and genome-wide analysis of a catalytically deficient SETDB1-like enzyme, MET-2, in Caenorhabditis elegans reveals that MET-2 promotes transcriptional silencing and fertility through both H3K9 methylation and focus formation, which blocks histone acetylation.

Mattout et al. show that the nuclear LSM2-8 complex acts with the exonuclease XRN-2 to degrade transcripts of H3K27me3-marked genes, thus facilitating gene silencing.

Establishment of H3K9-methylated heterochromatin and its functions in tissue differentiation and maintenance

Histone H3 Lys9 (H3K9)-methylated heterochromatin ensures transcriptional silencing of repetitive elements and genes, and its deregulation leads to impaired cell and tissue identity, premature aging and cancer. Recent studies in mammals clarified the roles H3K9-specific histone methyltransferases in ensuring transcriptional homeostasis during tissue differentiation.
