MLL oncoprotein levels influence leukemia lineage identities

The effects of chromosomal translocations involving the mixed-lineage leukemia (MLL) locus on gene expression regulation remain to be explored. Here, the authors find that MLL oncoproteins support lineage-switching events through dynamic chromatin binding.

Oncogenic Rag GTPase signalling enhances B cell activation and drives follicular lymphoma sensitive to pharmacological inhibition of mTOR

Some follicular B cell lymphomas harbour activating mutations in RRAGC, activator of the nutrient sensor mTORC1. Here the authors show that these mutations confer insensitivity to nutrient deprivation and synergize with paracrine cues from the supportive T cell microenvironment to accelerate lymphomagenesis, but impose vulnerability to inhibition of mTORC1.

They identify recurrent mutations in linker histone genes and genes involved in JAK-STAT signaling, NF-κB signaling and B cell development.
