MitoPerturb-Seq identifies gene-specific single-cell responses to mitochondrial DNA depletion and heteroplasmy

Burr and Auckland et. al develop MitoPerturb-Seq, which combines single-cell screening with multiomics to link nuclear genes to mitochondrial DNA (mtDNA) dynamics. They unveil core regulators of mtDNA copy number and characterize cell-cycle delays and transcriptional stress in response to mtDNA depletion.

Bcl-wav and the mitochondrial calcium uniporter drive gastrula morphogenesis in zebrafish

Ca²⁺ is an intracellular messenger that has a critical role in zebrafish development. Here Prudent et al. show that during gastrulation, the newly identified Bcl-2 homologue, Bcl-wav and the mitochondrial calcium uniporter regulate cell migration by controlling mitochondrial Ca²⁺storage.

Emerging mechanisms of genome degradation during the mtDNA life cycle

Mitochondrial DNA (mtDNA) is not static but undergoes a regulated ‘life cycle’ that encompasses replication, maintenance, spatial distribution and turnover. Here, we update emerging mtDNA degradative pathways, discuss the interplay between mtDNA quality control and heteroplasmy, and outline crucial questions that place the mtDNA life cycle at the forefront of mitochondrial biology.

An inherited mitochondrial DNA mutation remodels inflammatory cytokine responses in macrophages and in vivo in mice

Inherited mitochondrial DNA mutations can result in diverse clinical phenotypes. Here, the authors characterise a heteroplasmic tRNA^Ala mutation (m.5019A>G) in mice and demonstrate that macrophages carrying this mutation display altered function and metabolism in vitro, along with increased type I IFN release following LPS challenge in vivo.

SMARCA4/2 loss inhibits chemotherapy-induced apoptosis by restricting IP3R3-mediated Ca²⁺ flux to mitochondria

SMARCA4/2 loss in ovarian and lung cancers is associated with chemotherapy resistance. Here, the authors show that SMARCA4/2 deficiency in cancer cells reduces the expression of the ER-Ca²⁺ channel IP3R3 and subsequently calcium transfer to the mitochondria, which inhibits apoptotic cell death.

Oxygen tension modulates the mitochondrial genetic bottleneck and influences the segregation of a heteroplasmic mtDNA variant in vitro

Using an in vitro culture system, Pezet et al. studied the influence of oxygen on the mitochondrial DNA (mtDNA) in primordial germ cell-like cells (PGCLCs) in vitro. Low oxygen levels resembling in vivo reduced the cell mtDNA content causing a genetic bottleneck and the segregation of different mtDNA genotypes.
