Mechanisms of Disease: detrimental adrenergic signaling in acute decompensated heart failure

Little is known about the basic pathologic mechanisms in acute decompensated heart failure (ADHF), despite ADHF being a large clinical problem. Here, David Feldman, Terry Elton, Benjamin Sun, Mickey Martin and Mark Ziolo focus on β-adrenergic receptor signaling as one of the final common pathways that could directly contribute to ADHF.

Rationally engineered Troponin C modulates in vivo cardiac function and performance in health and disease

Heart contraction, which is decreased in disease, is determined by Ca²⁺binding to troponin C. Here, the authors combine a protein engineering approach with gene therapy to modulate heart contractility in mice with the use of rationally designed Troponin C variants, suggesting a new therapy for diseased hearts.

NF-κB inhibition rescues cardiac function by remodeling calcium genes in a Duchenne muscular dystrophy model

The molecular mechanisms leading to heart failure in patients with Duchenne muscular dystrophy are unclear. Here the authors show that NF-κB is activated in the heart of dystrophin-deficient mice and that its ablation rescues cardiac function through chromatin remodeling and activation of gene expression.

Advanced 4-chamber echocardiography techniques enable clinically matched precise characterization of heart disease progression in mice

Kacira et al. develop an apical 4-chamber-long-axis (A4CLAX) echocardiographic imaging approach for mice. They demonstrate that the A4CLAX method provides clinically comparable and comprehensive evaluation of cardiac function in murine models, with the sensitivity to detect subtle changes in cardiac performance during disease progression.

Exercise-induced 3′-sialyllactose in breast milk is a critical mediator to improve metabolic health and cardiac function in mouse offspring

Maternal exercise, before and during pregnancy, has a beneficial effect on offspring. Harris et al. report that exercise-induced release of oligosaccharide 3′-sialyllactose in mouse breast milk mediates the metabolic health benefits of maternal exercise on offspring by improving glucose and insulin tolerance and cardiac function.
