  • RORγt+ antigen-presenting cells (APC) are playing a role in the generation of oral tolerance via their interaction with regulatory T cells recognising dietary and commensal bacterial antigens. Here authors show that a cis transcriptional regulatory element is responsible for RORγt specifically in APCs through interaction with RUNX3 and formation of chromatin loops, which determines a transcriptional program that distinguishes these oral tolerance mediating APCs from RORγt + T cells.

  • Subsets of ILC3s upregulate the immunoregulatory checkpoint molecule CTLA-4 after stimulation in a microbiota-dependent manner, and advances to support CTLA-4+ ILC3s may represent a treatment opportunity in IL-23-driven chronic inflammation.

  • Recent studies have revealed a family of antigen-presenting cells (APCs) marked by the transcription factor RORγt that fundamentally shape immunity, inflammation and tolerance. This article reviews heterogeneity among RORγt+ APCs, their associated functions and the future promise of this new field.
