Necroptosis microenvironment directs lineage commitment in liver cancer

The tumour microenvironment determines which type of liver cancer develops, with transformed hepatocytes giving rise to intrahepatic cholangiocarcinoma or hepatocellular carcinoma depending or whether they are surrounded by cells undergoing necroptosis or apoptosis.

In this Review, Luedde and colleagues describe the complex role of regulated necrosis pathways, including necroptosis, pyroptosis and ferroptosis, in liver disease and cancer development. Potential therapeutic approaches modulating regulated cell death pathways in liver disease are explored.

A decision point between transdifferentiation and programmed cell death priming controls KRAS-dependent pancreatic cancer development

The involvement of cell death pathways in the early stage of pancreatic ductal adenocarcinoma (PDAC) development, especially KRAS-dependent acinar-to-ductal metaplasia (ADM), remains to be investigated. Here, the authors find that TAK1 mediates cell survival during ADM transdifferentiation through suppression of apoptosis and necroptosis, which could be targeted for prevention and treatment of PDAC.

The necroptosis-inducing kinase RIPK3 dampens adipose tissue inflammation and glucose intolerance

The kinase RIPK3 initiates necroptosis, which has been reported to promote inflammation in various pathological conditions. Here, the authors show that genetic ablation of Ripk3results in adipocyte apoptosis and white adipose tissue inflammation in obese mice, which promotes glucose intolerance.
