Through producing acetyl-CoA from acetate, ACSS2 epigenetically controls gene expression. However, whether and how ACSS2 regulates senescence remains unclear. Here, the authors link ACSS2-generated local acetyl-CoA to purine metabolism through PAICS acetylation that contributes to CCF-SASP.
