Irisin ameliorates obesity and insulin resistance via adipose tissue IL-33 and regulatory T cells

This study demonstrates that overexpression of irisin, an exercise-induced myokine, ameliorates obesity and insulin resistance in high-fat-diet-fed mice by increasing local IL-33 production and preserving ST2⁺ regulatory T cells in white adipose tissues.

A20 (TNFAIP3) deficiency in myeloid cells triggers erosive polyarthritis resembling rheumatoid arthritis

Geert van Loo, Rudi Beyaert, Dirk Elewaut and colleagues report that myeloid-specific deletion of Tnfaip3 in mice causes spontaneous destructive polyarthritis that resembles rheumatoid arthritis. Tnfaip3 encodes the A20 protein, which is involved in negative regulation of NF-κB signaling in response to proinflammatory stimuli.

A20 controls intestinal homeostasis through cell-specific activities

Aetiology of colitis is highly complex and incompletely understood. Here the authors show in mouse models that A20 deubiquitinase limits pro-inflammatory cytokine production in myeloid cells while inhibiting proapoptotic response to these cytokines in enterocytes, and that only upon losing both functions intestinal pathologies develop.

Elevated IL-33 levels have been correlated with metastasis and poor prognosis. Here the authors show in mouse tumour xenograft models that PDGF-BB produced by tumour cells induces IL-33 via Sox7 in tumour pericytes, and IL-33 promotes metastasis through its effects on tumour-associated macrophages.
