Synthesis of bioactive (1→6)-β-glucose branched poly-amido-saccharides that stimulate and induce M1 polarization in macrophages

Difficulty with purification, structural heterogenicity, and limited water solubility of β-glucans has significantly limited their therapeutic applications. Here, the authors report the synthesis of (1→6)-β-glucose-branched poly-amido-saccharides as glycan-mimetics and demonstrate macrophage stimulation and polarization.

In NAFLD, high levels of fatty acids in the liver impair lysosomal acidification. Here, the authors report the synthesis of novel biodegradable acid-activated acidifying nanoparticles that re-acidify lysosomes, restore autophagy, and reverse fasting hyperglycemia and hepatic steatosis in fat mice.
